Relationship of five aceh patchouli varieties (Pogostemon cablin (Blanco) Benth.) based on internal transcribed spacer (ITS) genes

Abstract


Patchouli plants are a class of essential oils that play an important role in increasing foreign exchange or increasing Gross Regional Domestic Product (GRDP) for countries used in the industrial world. Patchouli plants in Indonesia consist of three types of patchouli, namely Pogostemon cablin (Blanco) Benth. (patchouli aceh), Pogostemon hortensis Backer., (soap patchouli) and Pogostemon heyneanus Benth. (Java patchouli). Patchouli aceh (Pogostemon cablin (Blanco) Benth.) is the first type of patchouli plant to be widely cultivated in almost all regions of aceh, and has several varieties, including the sidikalang variety, tapaktuan variety, lhokseumawe variety, patchoulina 1 variety and patchoulina 2 variety. This study aims to determine the kinship of the five varieties because it is not yet known and there are no publications that discuss the kinship of the five varieties of aceh patchouli with a molecular approach using the ITS gene. The research method used is descriptive which includes several stages, namely the DNA extraction stage using the CTAB method, DNA amplification using the ITS gene, sequencing, alignment of sequencing data, and phylogenetic tree construction using MEGA X with the Maximum Parsimony method using a bootstrap of 1000 repetitions. The amplification results of 15 samples based on the ITS gene in this study produced DNA bands ranging from 650-700 bp. The results of the alignment were then compared with the data in GenBank. The results of the phylogenetic tree reconstruction based on the ITS gene obtained 2 groups forming monophyletic and polyphyletic. The first group SD (2), SD (1), TP (1), TP (2), TP (3), LH (1), LH (2), LH (3), P 1 (1), P 2 (1), P 2 (3) formed monophyletic in which the group members came from the same ancestor and the second group P 1 (2), P 1 (3), P 2 (2) have different DNA characters from the sidikalang, lhokseumawe and tapak tuan groups, so this group forms a polyphyletic group.
